Studies of Phytochemical Analysis, In vitro and In vivo Evaluation of the local Pseudoelephantopus spicatus Plant

The plant Pseudoelephantopus spicatus is used as folklore medicine in Bangladesh especially in rural side and Chittagong Hill tracts for appeasing many diseases. Methanolic extracts of plant of P. spicatus and its different Kupchan partitionates i.e. CH3OH soluble partition (MESP), n-C6H14 Soluble partition (HSP), CH2Cl2 Soluble partition (DCMSP), Ethyl acetate Soluble partition (EtAcSP) and Aqueous Soluble partition (AQSP) were subjected to assay for in vitro and in vivo investigations. These are rudimentary for searching the consistency and potency of plant extracts. The result of bioassays is impactful, since these preclinical trials extrapolates a valid method which is being used for drug development. Qualitative phytochemical screening showed the presence of Tannin, Alkaloid, Flavonoid and Steroid. In investigations of total phenolic contents, the EtAcSP showed highest presence of phenolic compounds (102.56mg of GAE /gm of extractives) among the partitionates, the MESP of plant of P. spicatus showed the largest free radical scavenging activity having the value of IC50=76.11?g/ml on 2,2-diphenyl-picrylhydrazyl (DPPH), kupchan fractions were taken to antimicrobial screening with disc diffusion method showed DCMSP of methanolic extract of P. spicatus exhibited good inhibition against growth of bacteria having zone of diffusion of inhibition ranged from 6-16 mm, the EtAcSP of plant of P. spicatus exhibited highest thrombolytic activity (290.16%), the brine shrimp lethality was given by DCMSP (1.99?g/ml) subsequent by HSP (2.03?g/ml), AQSP (2.05?g/ml), MESP (2.50?g/ml) and EASP (2.60?g/ml). The herb is less toxic. The other peripheral and central analgesic activity, antidiarrheal, antidepressant and hypoglycemic activity assay exhibited encouraging results at both doses of 400- and 200- mg/kg body weight in Swiss-Albino mice model. In Fatty acid analysis Hexane extract of plant of P. spicatus contains highest proportion of bound fatty acid of cis-9-oleic acid (34.14 %) and free fatty acid of Palmitoleic acid (12.25 %). This finding indicates that unsaturated fatty acids are higher in proportion as bound fatty acids (BFAs) than that of free fatty acids (FFAs). Therefore, regarding the potential bioactivity, the plant P.spicatus can further be used for studying widely to find out their unrevealed efficacy and to generalize their uses as folklore medicines.
