Isolation of a novel strain Bacillus sp.in producing neutral and heat stable beta-1,4 endoglucanase activity

Rahman Laibi Chelab, Dan Dan Wang and Xingyong Yang

Neutral a β-1-4 - endoglucanase was produced by aerobic bacteria a newly strain Bacillus sp .RL. Isolated from soil, Chong Qing province, China. In the present study the bacteria produce a significant amount of enzymes after 72 hrs of fermentation CMC medium were 0.1852 U/ml of endoglucanase, 0.5814 U/ml of FPase activity and 2.9940 mg/ml of extracellular protein. These strains showed heavy growth and liquefaction of 7.0, 35°C. The endoglucanase retained 85.5% and 66.9% of original activity at 90°C and pH 10, respectively. The endoglucanase activity was inhibited by CuSO4.4H2O (54%), while the best sulfate source improved enzyme activityFeSO4.7H2O (275%). FPase was markedly activated by CuSO4.4H2O> ZnSO4.2H2O> FeSO4.7H2O> Na2SO3, 180%,158%,152% and119% respectively.
